Hemodynamic and electromechanical effects of paraquat in rat heart

Paraquat (PQ) is a highly lethal herbicide. Ingestion of large quantities PQ usually results in cardiovascular collapse and eventual mortality. Recent pieces evidence indicate possible involvement oxidative stress- inflammation-related factors PQ-induced cardiac toxicity. However, little information exists on the relationship between hemodynamic electromechanical effects involved acute poisoning. The present study investigated exposure hemodynamics electrocardiogram (ECG) vivo , left ventricular (LV) pressure isolated hearts, as well contractile intracellular Ca 2+ properties ionic currents myocytes rat model. In anesthetized rats, intravenous administration (100 or 180 mg/kg) induced dose-dependent decreases heart rate, blood pressure, contractility (LV +d P /d t max ). Furthermore, prolonged PR, QRS, QT, rate-corrected QT (QTc) intervals. Langendorff-perfused (33 60 μM) decreased LV (10–60 reduced amplitudes transients fractional cell shortening concentration-dependent manner myocytes. Moreover, whole-cell patch-clamp experiments demonstrated that current amplitude availability transient outward K + channel ( I to ) altered its gating kinetics. These suggest cardiotoxicity mainly from diminished inhibited channels cardiomyocytes, which lead force suppression QTc interval prolongation. findings should provide novel cues understand electrical disturbances may aid development new treatment modalities.
